I would assume the answer is Yes as third-party flight trackers certainly "know" ahead of time and the gate monitors in the terminal reflect this.
For example, if one is waiting at the gate for a flight that is beginning its flight journey under its flight number but the inbound plane hasn't arrived yet, one can log into FlightAware and track the assigned inbound physical aircraft even though it would be operating under a different flight number.
